Hotel room size vary for simple (and small) to lavish, multilevel penthouse suites (some with their private elevator), so there is no standard. But a 50 story hotel would normally be expected to be in an area where land is expensive, and the hotel would then cater to affluent patrons who expect and are willing to pay for large accommodations.
How big the floor plan depends on how many rooms you need, how big you want each to be, and so on. Check out the SkyScraper site (see link) that will give you an idea of how wide building are as a function of how tall they are. But also how much variation there is.
a hotel room should probably be around 200-300 sqft. for the base of the hotel, you need to calculate that.
